From its sites in Cambridge, Massachusetts and Zurich, Switzerland, Repertoire’s team is creating a new pipeline of therapeutic immune medicines enabled from its DECODETM discovery platform. Repertoire is currently using its proprietary platform to discover novel epitopes and TCRs from patients with autoimmune diseases such as type 1 diabetes and multiple sclerosis, creating programmable tolerizing mRNA vaccines to restore immune homeostasis in these indications. In oncology, Repertoire’s decoding of tumor-infiltrating lymphocytes and the antigens they target is enabling development of TCR bispecific therapies and mRNA cancer vaccines to treat solid tumors.

Role Overview

We are seeking an enthusiastic Research Associate I/II (RA) to join our Platform Discovery Team. Some of the primary responsibilities of the RA will include preparation of reagents used in our DECODE assays, quality control of materials produced, and execution of next-generation sequencing experiments. These activities will be performed both manually and with the help of liquid handling instrumentation. The RA will work collaboratively as part of a cross-functional team of immunologists, molecular biologists, and protein scientists to successfully execute experiments and contribute to process optimization. We are looking for a team player who will thrive in a fast-paced and dynamic environment.

Key Responsibilities

  • Generate, purify, QC and track in-house materials required for the execution of our DECODE platform.
  • Prepare reagents at the bench with manual pipetting.
  • Operate liquid handling robots to produce materials at high throughput/complexity.
  • Work closely with scientific program teams to execute single-cell sequencing or bulk sequencing experiments to meet project objectives.
  • Isolate DNA/RNA from primary human samples (blood and tissue) and prepare next-generation sequencing libraries for bulk and single-cell measurements.
  • Perform quality control techniques, such as qPCR and gel electrophoresis, on nucleic acid samples to ensure high quality data generation.
  • Perform routine maintenance on highly specialized laboratory equipment, including next-generation sequencing instruments.
  • Assess process outputs in collaboration with the Platform Team as well as analyze and present experimental results at department team meetings.
  • Order and keep inventory of critical reagents to ensure material availability to meet project timelines.

Qualifications/Experience

  • Bachelor’s or Master’s degree (or equivalent) in scientific discipline or a health-related field.
  • Up to 2+ years post B.S. experience in relevant experimental research area in industry or academia (career level to be determined based on experience).
  • Strong background in molecular biology and associated general laboratory techniques (PCR, qPCR, ddPCR, nucleic acid purification),.
  • Familiarity with single cell methods (eg. 10X Genomics, Smart-Seq).
  • Familiarity with liquid handling instrumentation is preferred (E.g. Hamilton, Tecan).
  • Ability to prioritize and complete tasks in a timely manner.
  • Cursory knowledge of genetics, immunology, and cancer biology.
  • Ability to think critically and perform functions with minimal guidance.
  • Impeccable documentation practices and attention to detail.
  • Excellent written and verbal communication skills.
